Amnis Image Stream Mark II
The Amnis ImageStreamX Mark II imaging cytometer combines the fluidic sample delivery of flow cytometry with fluorescence microscopy imaging to rapidly capture hundreds of digital fluorescence images per sample, and to analyze populations with flow cytometry-like plots and gates. This hybrid technology is especially well-suited to measure statistics on sample populations that are defined in part by morphology, such as assaying nuclear factor kappa B (NF-kB) translocation from the cytoplasm to the nucleus.
The ImageStream features 405 nm, 488 nm, and 642 nm collinear lasers and six fluorescence detectors (457/45, 533/55, 577/35, 610/30, 702/85, 762/35). The manufacturer's suggested fluorochrome combination for our configuration is: DAPI, AF488, PE, PE-TexasRed, AF647, and dark field (SSC). Bright field (BF) imaging can replace any detector. (If the 610/30 detector is chosen for BF the panel can be used in parallel without modification for cell sorting on the SH800.) Available magnifications are 20x, 40x, or 60x.
